Autolytic mechanism for spheroplast formation in Bacillus cereus and Escherichia coli. J. Bacteriol. 90:1355-1364. 1965.-Spheroplasts of Bacillus cereus strain T and Escherichia coli B were prepared by incubating early log-phase cells in appropriate buffers and stabilizers for 3 hr at 30 and 37 C, respectively. Upon incubation in 0.05 m tris(hydroxymethyl)aminomethane buffer osmotically stabilized with 16% polyethylene glycol at pH 7.5, 99% of the B. cereus cells formed spheroplasts; 90% of the E. coli cells were converted to spheroplasts in 0.4 m sodium acetate buffer osmotically stabilized with 1.6 m sucrose at pH 6.0. The extent of spheroplast formation was determined by phase-contrast microscopic examination, by measuring the rate of fall of optical density in the reaction mixture when subjected to osmotic shock, and by viable intact cell counts. The effect of a selected group of metabolic inhibitors on the autolytic system of B. cereus and E. coli has been examined. B. cereus and E. coli wall components comprising 26% of the dry weight of the original cellular material were recovered from dialyzed fractions by precipitation in 70% ethyl alcohol. Chemical and chromatographic analysis of cell-wall hydrolysates from B. cereus and E. coli indicated the presence of glucosamine, alanine, lysine, glycine, aspartic acid, diaminopimelic acid, glutamic acid, and muramic acid.

Identification of Anise (Pimpinella anisum L.) Essential Oil Compounds and Investigation of its Effect on Some Foodborne Pathogens: Bacillus cereus, Staphylococcus aureus, Escherichia coli O157: H7and Salmonella typhimurium
Background and Objectives: Nowadays, use of essential oils in food preservation is popular by researchers not only as antimicrobial agents but also as replacements for synthetic harmful preservatives. Objectives of this study were extraction and identification of Anise essential oil (AEO) compounds as well as assessment of the oil antimicrobial properties on four Gram-negative and Gram-positive...
متن کاملThe Survey on Antimicrobial Effects of Methanolic Extract of Carum Copticum L. on Staphylococcus Aureus, Bacillus Cereus, Escherichia Coli and Pseudomonas Aeruginosa in Laboratory Conditions
Introduction: Nowadays, the prevalence of drug resistance to synthetic drugs is increasing in many countries in the world; therefore, many efforts have been made to find new compounds as an appropriate alternative to antibiotics. This study was performed to investigate the antibacterial effect of methanolic extract of Carum copticum L. on some pathogenic bacteria. Methods and Materials: In thi...
متن کاملThe autolytic phenotype of the Bacillus cereus group.
AIM To determine the autolytic phenotype of five species in the Bacillus cereus group. METHODS AND RESULTS The autolytic rate of 96 strains belonging to five species in the B. cereus group was examined under starvation conditions at pH 6, 6.5 and 8.5 in different buffers. The autolytic rate was strain-dependent with a wide variability at pH 6, but higher and more uniform at pH 6.5. At pH 8.5,...

متن کاملEffects of Aronia melanocarpa constituents on biofilm formation of Escherichia coli and Bacillus cereus.
Many bacteria growing on surfaces form biofilms. Adaptive and genetic changes of the microorganisms in this structure make them resistant to antimicrobial agents. Biofilm-forming organisms on medical devices can pose serious threats to human health. Thus, there is a need for novel prevention and treatment strategies. This study aimed to evaluate the ability of Aronia melanocarpa extracts, subfr...
